Research suggests that Lion's Mane mushroom may offer some support for depression and mood, with evidence drawn from a range of study types including randomized controlled trials, animal and in vitro studies, and multiple narrative and systematic reviews. The most directly relevant human evidence includes a 2010 RCT reporting reduced depression and anxiety scores in female participants after four weeks of use, and a 2023 pilot RCT in healthy young adults that found a trend toward reduced subjective stress after 28 days, though that study was small and authors cautioned the findings are preliminary. Preclinical research and mechanistic reviews consistently point to plausible pathways — including stimulation of nerve growth factor and BDNF, modulation of neurotransmitter systems, anti-inflammatory activity, and gut-brain axis effects — that could underlie antidepressant-like actions observed in animal models. Overall, the body of evidence leans in a supportive direction, but the human clinical data remain limited in sample size and scope, meaning firm conclusions about efficacy for depression cannot yet be drawn and further well-controlled trials are needed.
