Research suggests that L-glutamine plays a meaningful role in supporting intestinal barrier function, gut immune regulation, and gut microbiota development, with the most consistent evidence coming from animal studies — including a systematic review of piglet research and multiple livestock-focused investigations — showing improvements in gut structure, reduced inflammation, and enhanced early-life gut health when L-glutamine is supplied through diet or maternal sources such as milk. Studies indicate that L-glutamine's benefits in the gut may include modulating inflammatory signaling pathways, supporting intestinal tissue architecture, and contributing to beneficial shifts in gut bacterial communities, with one study identifying increased milk L-glutamine as a key protective factor against neonatal intestinal inflammation. However, the evidence is not uniformly positive: one study found that L-glutamine triggered mast cell degranulation and release of inflammatory mediators in intestinal tissue during fat absorption, suggesting the supplement may also activate immune responses in ways that are not fully understood and could warrant caution. The overall body of evidence reviewed here is composed largely of animal studies across livestock and fish models, with limited direct human research, meaning the applicability of these findings to human gut health remains uncertain and the field would benefit substantially from well-designed human clinical trials.
