Research suggests that bitter compounds may influence appetite regulation by activating bitter taste receptors found throughout the gastrointestinal tract, triggering the release of hormones such as GLP-1, CCK, PYY, and GDF15 that signal fullness and suppress hunger. The body of evidence includes numerous mechanistic reviews, two randomized controlled trials, and two systematic reviews, and the majority of this literature leans supportive of the proposed mechanism — particularly at the preclinical level, where cell and animal studies consistently show effects on gut hormone release, gastric motility, and food intake. However, the two systematic reviews offer an important counterbalance, finding that results across human studies are largely inconsistent, that most individual trials show no significant changes in appetite or food intake, and that only a few hormones such as motilin and ghrelin respond with any reliability to bitter compounds in humans. Studies indicate that while targeting intestinal bitter taste receptors remains a scientifically plausible and actively investigated strategy, the current human evidence is insufficient to firmly establish that digestive bitters meaningfully reduce appetite or food intake, and much of the optimism in this area still rests on preclinical findings that have not yet been reliably replicated in clinical settings.
