Research suggests that 5-HTP may have modest sleep-promoting effects, though the evidence base in humans remains limited. The two randomized controlled trials in this set both point in a supportive direction: one found that 5-HTP supplementation improved subjective sleep quality in older adults who were poor sleepers at baseline, while the other observed increased REM sleep percentage in Parkinson's disease patients, though both trials were small and short in duration. Several animal studies further support the biological plausibility of 5-HTP's role in sleep, particularly when combined with GABA, where synergistic effects on sleep onset and duration were observed in mice, rats, and fruit flies. However, the majority of the evidence comes from preclinical models or studies focused on other compounds that happen to involve serotonergic pathways, and the few human trials available are limited by small sample sizes, brief intervention periods, and modest doses, so larger and longer clinical trials are needed before firm conclusions can be drawn about 5-HTP's effectiveness for improving sleep quality in the general population.
